Supplementary information

FGF21 (Fibroblast Growth Factor 21) is a metabolic hormone that plays an important role in regulating energy balance, glucose metabolism, and lipid metabolism.

FGF21 is mainly produced by the liver. Its production is stimulated by metabolic stress, such as fasting, ketogenic diets, cold exposure, and high-fat diets.

FGF21 exerts its effects by binding to FGF receptors (FGFRs) together with the co-receptor β-Klotho, which is highly expressed in metabolic tissues such as liver, adipose tissue, and the central nervous system. Activation of this receptor complex triggers intracellular signaling pathways, including MAPK/ERK signaling, leading to metabolic regulatory effects.

Biologically, FGF21 promotes glucose uptake, fatty acid oxidation, and energy expenditure, while also improving insulin sensitivity.
