information
α-Synuclein
is a presynaptic neuronal protein involved in synaptic vesicle trafficking and
neurotransmitter release. In Parkinson’s disease and related synucleinopathies,
α-synuclein undergoes misfolding and aggregation, leading to the formation of
Lewy bodies, a pathological hallmark of the disease.
Several
α-synuclein species are commonly measured as biomarkers in research and
clinical studies, including total α-synuclein, oligomeric α-synuclein, and
phosphorylated α-synuclein. Total α-synuclein reflects the overall protein
level, while oligomeric α-synuclein represents aggregated soluble forms that
are considered the most neurotoxic species and are believed to play a key role
in neuronal damage. In addition, phosphorylated α-synuclein at serine 129 is
strongly associated with pathological aggregation, as the majority of
α-synuclein present in Lewy bodies is phosphorylated at this residue.
Measurement
of these different α-synuclein species helps characterize disease pathology,
progression, and therapeutic response in Parkinson’s disease studies.
